Objective: This study assessed the in vitro enzyme-inhibitory potential of Taraxerol acetate against Lipoxygenase and Cyclooxygenase Isoforms, along with in vivo and in vitro toxicity. It aims to establish the anti-inflammatory properties of Taraxerol Acetate (TA).
Methods: We assessed lipoxygenase inhibition by spectrophotometric monitoring of hydroperoxide production from linoleic acid. Cyclooxygenase-1 and Cyclooxygenase-2 inhibition was assessed by radioimmunoassay of prostaglandin E₂ production. Acute in vivo toxicity was evaluated in mice after intraperitoneal administration of TA at doses of 100, 200, and 400 mg/kg body weight, with observation over fourteen days. In vitro cytotoxicity was assessed using the MTT assay in LCKM-2 monkey kidney epithelial cells and mouse hepatocytes.
Results: TA demonstrated significant LOX inhibition with an IC₅₀ of 82.19 ± 0.01 μM, compared with the standard inhibitors baicalein (IC₅₀ = 22.0 ± 0.05 μM) and tenidap sodium (IC₅₀ = 41.6 ± 0.02 μM). TA showed IC₅₀ values of 116.3 ± 0.03 μM for cyclooxygenase 1 and 94.7 ± 0.02 μM for cyclooxygenase 2. The standard ibuprofen showed IC₅₀ values of 14.91 ± 0.06 µM and 27.20 ± 0.05 µM for cyclooxygenase-1 and cyclooxygenase-2, respectively.
Conclusion: TA showed moderate inhibitory activity against lipoxygenase and cyclooxygenase enzymes.
